Use of biological properties of plant viruses for their detection
Biological tests are based on the use of indicator host(s) expressing a range of symptoms according to the differential properties of the inoculated isolate. Applied to PVY isolates, such viral detection/characterisation approach is mainly performed using Nicotiana tabacum cv. Xanthi (susceptible host to necrotic properties of PVYN, PVYNTN and PVYN-W isolates), and selected varieties of potato (e.g. Nicola expressing hypersensitive reaction to PVYO and producing tubers with necroses symptoms when infected by PVYNTN isolates). Chenopodium amaranticolor makes possible the distinction between PVYO (local lesions on inoculated leaf) and PVYN (systemic infection). Other indicator hosts exist and could be used to identify PVY groups and/or variants.
